The achievement of the Molecular & Microbial Engineering Group, "Research on the Function and Catalytic Mechanism of Biomass Polysaccharide Degrading Enzymes", completed by Fuli Li, Ming Lu, Hang Su, Xiaoqing Ma, Kundi Zhang, has won the second prize of Shandong Provincial Natural Science Award. Director of QIBEBT, Zhongmin Liu, has commended and awarded the group at the annual working meeting.

The team focuses on the function and catalytic mechanism of polysaccharide degrading enzyme, which are the key factors of biomass conversion. Starting from thermophilic microorganisms, the research team deeply analyzes the enzymatic performance and catalytic mechanism of the polysaccharide degrading enzyme, as well as the relationship between thermal stability and environmental adaptive evolution, thereby recognizes and understands the coevolutionary mechanism between environmental microorganisms and biomass, and effectively solves the problems of high cost and low efficiency of enzyme preparation. The main achievements achieved by the team are as follows. (1) A "wire walking" model of sustainable endocellulase is proposed and has avoided the "inactivation" of fungal derived enzymes. (2) A new mechanism of substrate selectivity and catalysis of thermophilic polysaccharide degrading enzymes GH5 family bifunctional cellulase/lichanase are proposed. A number of thermostable alginate lyases are screened and characterized, and the relationship between the metal ion binding power and the mechanism of thermal adaptation has been revealed. (3) High yield heterologous expression strain of alginate lyase has been constructed. Through the optimization of enzyme preparation, an efficient catalytic system of seaweed oligosaccharide is constructed.

The projector leader, researcher Fuli Li, has cooperated with many internationally renowned research groups, presided over the "Sino-German Center for Research Promotion" project of the National Natural Science Foundation of China, and jointly published articles with many well-known professors, like Professor Robert Kelly of the University of North Carolina in the United States, and Professor Edward Bayer of the Weizmann Institute of Science in Israel. He also presided over the compilation of Thermophilic Microorganisms (published in 2015). Furthermore, the high-temperature and high-efficient enzymatic hydrolysis process of marine oligosaccharides developed by the team has already been industrialized in Weifang Macroalga and Fujian Lifecome Biochemistry Co., Ltd.  (Provided by Xiaoqing Ma)
